| Outline of Annual Research Achievements |
ホスファチジルコリン多様性生合成メカニズムの一端を報告した。リン脂質は従来はde novo経路により作られたものが、remodeling経路で多価不飽和脂肪酸が導入されるというのが定説であった。18:2や22:6の脂肪酸はde novo経路のホスファチジン酸生合成の段階でリン脂質に組み込まれホスファチジルコリン組成に影響することがわかった。16:0や18:1の脂肪酸はremodeling経路のホスファチジルコリン生合成の段階で組み込まれやすいことがわかった。また、ホスファチジルコリン生合成酵素の一つであるLPCAT1の欠損マウスは過換気呼吸モデルで致死率が高いことがわかった。これは肺サーファクタント脂質の質が変化することにより、肺にダーメジがあり、炎症を誘発したためであった(Harayama et al. Cell Metab. 2014)。アラキドン酸の導入の仕組みについても新たな知見があった(Hashidate et al. eLife, 2015)。
血小板活性化因子（PAF）生合成酵素であるLPCAT2の生化学的な解析が進んだ。LPCAT2は、PAFやATPによる刺激３０秒程度で、３４番目のSerのリン酸化によって活性化された。これはCa/PLC/cPKC経路によるものであった。マクロファージにおいてLPCAT2の３種類の調節機構が解明された（Morimoto et al J Biol Chem. 2014）。さらにLPCAT2阻害剤スクリーニングを１７万化合物から行った。LPCAT2阻害剤としてTSI-01を報告した(Tarui et al. J Lipid Res. 2014)。
１５０種程度のエイコサノイド一斉定量方法を開発した。同時解析が可能となる（Yamada et al J. Chrom. B, in press）。

Reason
ホスファチジルコリン生合成メカニズムの一端の解明とLPCAT2の酵素学的な解析を行えた。また、LPCAT1欠損マウスの正常肺機能のための重要性も発表できた。さらに、エイコサノイド測定方法も樹立した。よって、おおむね順調である。
他のマウスに関しては引き続き継続している。

| Strategy for Future Research Activity |
引き続き酵素欠損マウスを用いたリン脂質代謝解析と細胞機能、疾患との関わりを解明する。
LPCAT1欠損マウス：肺疾患、網膜疾患、LPCAT2欠損マウス：血小板活性化因子（PAF）関連炎症モデル実験、LPCAT3欠損マウス：小腸、肝機能。誘導的ノックアウトの解析
また、生化学レベルでは阻害剤開発、細胞レベルでは分化によるリン脂質、生合成酵素、脂肪酸組成の違いをプロファイリングし、機能との関連を調べる。
