Outline of Annual Research Achievements

膜リン脂質多様性の生物学（いかに多様性を樹立するか、またその生物学的意義は何か）について、平成２７年度は以下の様な成果を上げている。第一は膜アラキドン酸含有リン脂質が私共の単離したLPCAT3という酵素により行われること、また、この酵素を欠損し、アラキドン酸含有リン脂質の低下した動物は小腸や肝にトリグリセリドが蓄積し、正常なリポタンパクの合成が阻害されることを報告した。この際、プロスタグランディンなどエイコサノイドの産生には影響を与えなかった。すなわち、脂質二重膜のアラキドン酸は単にエイコサノイドの前駆体として働くだけではなく、膜の曲率を形成し、小胞体でのトリグリセリド輸送に影響を与えることを示した。この研究と並行し、脳ではホスホリパーゼA2ではなく、モノアシルグリセロールリパーゼの働きでアラキドン酸が放出され、発熱その他の神経作用に関係することも明らかにした。１５０種に及びエイコサノイドの一斉定量系の確立、肺サーファクタント分子の解析などの新たな方法を開発し、発表した。

Reason

研究は予定通りに進んでいる。DHAなどの多価不飽和脂肪酸の導入メカニズムや、関連酵素欠損マウスの表現形の解析などをさらに加速する必要がある。また、確立したエイコサノイド一斉定量系を用い、血液や尿などの液体サンプルでの解析が課題として残っている。

Strategy for Future Research Activity

現在、作成中の各種変異マウスの膜リン脂質の解析、表現形との関係を明らかにする。さらに、ヒト検体の解析などにより、膜リン脂質の変化、代謝物の変化が疾患との関係を総合的に明らかにする。
