Outline of Annual Research Achievements

■産業動物は、ヒトと同じく多くの環境化学物質に曝露されており、化学物質による急性中毒や慢性曝露による病態が報告されている。産業動物の化学物質汚染は食の安全に直結する重要な問題でもあるが、一方で、ヒトや実験動物以外で化学物質の感受性の種差や個体差に関する研究は極めて少なく、これがケミカルハザードを引き起こす大きな原因の一つとなっている。
■本研究では、国内外の産業動物種が有する化学物質への生体防御機構と、化学物質感受性の種差や個体差を引き起こしている要因を明らかにし、研究基盤を構築することを目的押した。
■今年度は哺乳類の持つシトクロムP450、グルクロン酸抱合酵素、硫酸抱合酵素の代謝活性の動物種差を明らかにした。また、ピレンをモデル化合物とし、尿を用いた簡便な第II相反応のスクリーニング方法を確立した。さらに、シンテニー解析や偽遺伝子化のスクリーニングから、第II相反応の脆弱な動物種について報告した。
■フィールド調査により、各動物種に蓄積する環境汚染物質の分析を重金属を中心に行い、その汚染源について安定同位体などを用いて解析を行った。

Reason

■動物種差の比較データについて、順調に研究基盤を構築しており、学会発表や国際学術雑誌にへの投稿も行うことができた。これまで行ってきた、異物代謝酵素類の動物種差に関する研究は、いくつかの論文に掲載することができ、初めて哺乳類の化学物質に対する防御機構の種差を広範の生物種で明らかにして報告することができた。
■また、フィールドにおいて家畜（ウシ、ヤギ、ブタ、ヒツジ、ニワトリ）が暴露される化学物質の同定も進めており、有機ハロゲン化合物、重金属類などについて、その汚染源も含めた解析を進めることができている。

Strategy for Future Research Activity

■計画に変更はない。
■動物種差をよりを明らかにするために再構成系の実験を行い、各種酵素のクローン化を進める。家畜に蓄積する環境汚染物質の化学分析を引き続き行い、その汚染源について明らかにする。また、重金属についてはフィールドにおいても曝露実験を行い、濃度や安定同位体解析から、家畜における生体内動態を明らかにする。
■国際学術雑誌への研究成果の報告や学会において成果発表を行う予定であるが、さらに、アウトリーチ活動としてWEBサイトを用いた研究成果の発信や、高校生へ野指導などを通じて研究活動を紹介するなど、社会への貢献や情報発信も積極的に行う予定である。
