免疫グロブリン様受容体の炎症性疾患における病理学的意義の解明

Outline of Annual Research Achievements

MAIR-I、MAIR-IIは骨髄球系細胞に発現するIgLRであるが、MAIR-I遺伝子欠損マウスにおいて、OVA蛋白とAlumによる免疫で誘導されるアレルギー性気道炎症が野生型マウスと比較し、亢進することを示した。これは、MAIR-I遺伝子欠損マウスにおいてAlumによって誘導されたアポトーシス細胞を炎症性樹状細胞が認識し、Th２応答を誘導することによるものであることを明らかにした (J Immunol, in press)。
MIAR-II遺伝子欠損マウスでは、盲腸結紮後穿孔(Cecum ligation and puncture; CLP)により腹膜炎から敗血症を誘導すると、野生型マウスと比較し、有意に生存期間が短縮することを見いだした。炎症性単球に発現するMAIR-IIがLPS刺激によって炎症性単球の接着分子VLA4の活性化を誘導し、感染局所への遊走を促進していることを明らかにした (Nat Commun, 2014)。
