Outline of Annual Research Achievements

ヒトiPS細胞から誘導したミエロイド系血液細胞に、細胞増殖因子（cMYC + BMI1等）をコードする遺伝子を導入して発現させ、増殖性ミエロイド細胞（iPS-ML）を誘導することが出来た。さらにiPS-MLにGM-CSFとIL-4を添加して培養することにより樹状細胞（iPS-ML-DC）を誘導し、アロHLAクラスI 反応性CTLから逃避できるTAP欠損iPS-ML-DCについても樹立できた。このようにして作製された iPS-ML-DCが、我々がこれまでに同定したCTLやTh1細胞の活性化を誘導する癌抗原ペプチドを当該抗原に特異的なT細胞に提示して、これらを活性化できることを証明した。
またiPS-MLおよびiPS-ML由来の樹状細胞を用いた癌免疫療法の実用化をめざして、学内に設置されたCPC (Cell Processing Center)内の、GMP準拠培養によるiPS-MLの大量生産システムを利用して、まずヒトiPS-MLならびにiPS-ML-DCの大量生産が可能か否かについて検証中である。
以上の人工的な樹状細胞の大量培養系を簡略化すると共に、迅速かつ腫瘍化する危険性が少ない樹状細胞の大量培養を目指して、ヒトiPS細胞からではなくヒト末梢血中のCD14陽性単球に、細胞増殖因子（cMYC + BMI1等）をコードする遺伝子を導入して発現させ、増殖性ミエロイド細胞（CD14-ML）を分化誘導することに成功した。このCD14-MLから誘導した樹状細胞が、抗原のプロセッシングならびにT細胞への抗原提示機能を有することを証明した。
