Outline of Annual Research Achievements

Raffaelea quercivoraの宿主樹木に対する病原力は菌株間で差異があると示唆された.本研究では,本菌の病原力と材内での菌糸伸展との関連,その菌糸伸展に関与する樹木細胞壁分解に関わる酵素群を明らかにすることを目的とした.ミズナラの成木の枝と苗木にそれぞれ本菌4菌株の接種を行い,枯死率と非通水域,材内における菌糸伸長量を測定した.また,同4菌株のミズナラ材粉末を添加した培地上での菌糸伸長量と12種類の発色酵素基質を用いてそれらの酵素活性を評価した.3菌株の枯死率と非通水域の割合,材内における菌糸伸長量は対照木に比べて有意に大きく,非通水域の割合と菌糸伸長量との間で有意な正の相関が認められた.同3菌株の材添加培地上での菌糸伸長量は他の1菌株よりも有意に大きかった.さらに,その両者でペクチンとヘミセルロースの分解に関わる4種類の酵素活性が異なっていた.以上より,本菌の病原力の差異には材内における菌糸伸展が関与し,その伸展にはペクチンとヘミセルロース分解に関わる酵素活性が関与する可能性が指摘された.
