生体イメージングを活用した好塩基球の動態・機能の解明

Research Abstract

鉤虫（皮膚から侵入し、肺を通過して、最終的に消化管に寄生する寄生虫）に１度感染すると、宿主動物は寄生虫感染に対する免疫（耐性）を獲得し、その結果２度目以降の感染では寄生虫排除が効率良くおこることが知られている。しかし、耐性がどのようにしておこるのかは不明であった。私たちは、独自に開発した好塩基球特異的に緑色蛍光色素蛋白質を発現させた遺伝子改変マウスの生体イメージング解析から、２度目の鉤虫感染では皮膚の感染部位に好塩基球が集積していることを見いだした。２度目の感染では、寄生虫が侵入した皮膚局所に多数の寄生虫が捕捉されて肺・消化管への移行が阻止されていたが、誘導型好塩基球欠損マウスを用いて感染前に好塩基球を除去すると皮膚での寄生虫捕捉が解除され、感染が肺へと拡大した。すなわち、好塩基球が鉤虫に対する獲得免疫（耐性）に重要な働きをしていることが明らかとなった。さらなる解析から、皮膚感染部位に集積した好塩基球が産生するIL-4が単球・マクロファージに作用してM2型マクロファージの生成を促進すること、M2型マクロファージの産生するarginase 1が皮膚での寄生虫捕捉に寄与していることが判明した。これらの発見は、寄生虫感染に対する効率良いワクチンを開発する上で極めて有用であると考えられる。寄生虫感染局所やアレルギー炎症部位に遊走してくる好塩基球が実際に活性化する様子を可視化する目的で、好塩基球内のCa2+濃度の上昇あるいは脱顆粒にともなう分泌顆粒内pHの上昇にともなって蛍光を発するインディケーター蛋白質の発現コンストラクトを作製した。これらの組換遺伝子を好塩基球に発現させたところ、細胞活性化に伴って蛍光を発することが確認できた。現在、生体内での好塩基球活性化モニターに向けた条件検討を進めている。

Strategy for Future Research Activity

生体イメージングを応用した好塩基球の遊走・集積・機能解析をさらに発展させるとともに、寄生虫感染局所やアレルギー炎症部位に集積した好塩基球の活性化を可視化・モニターするシステムを樹立し、種々の生体反応における好塩基球の役割を解明する。

Expenditure Plans for the Next FY Research Funding

年度末の３月中に、出張予定が複数入り、主にその旅費予算用として、また、年度末にも、緊急に入手の必要な研究のための物品等の購入が可能となるよう、必要と見込まれる程度の金額を未使用のまま残したため。
主に、海外出張旅費（事務局から支給される補助では不足する航空券代の一部等）、および国内出張旅費（研究代表者本人および、研究代表者の所属、代表する研究室の研究成果発表者の学会出張にかかる、旅費、経費等）として支出する。また、研究遂行のための試薬、プラスチック製品、実験動物等、を緊急に入手したい場合の物品費として使用する。
