| Outline of Annual Research Achievements |
顎関節症に伴う慢性筋痛の多くはその発症メカニズムは不明であり，治療法も未だ確立されていない．近年，筋組織の治癒過程におけるマクロファージ（Mφ）の重要性が報告されている．そこで，慢性筋痛の新規治療開発を目的として，マウス咬筋部の疲労回復，および治癒過程におけるMφの役割と賦活化のメカニズムについて明らかとするため，マウス（Balb/cマウス，オス，5週齢）を用い，その咬筋部に咀嚼様運動に伴う筋疲労を誘導し，組織内におけるMφの動態とIL-1の発現動態について検討を行った．加えて，Mφ欠損により，筋疲労の回復遅延が生じることを明らかとする
リポゾーム封入クロドロネート（Clo-Lip）の静脈内投与（0.2ml/mouse）により組織内のMφが枯渇する．Clo-Lip投与による祖写経運動の持続量に有意な減少が示され，主要なマイオカインであるIL-1βおよびIL-6の発現量の低下が示された．IL1欠損マウスにおけえるIL-6誘導の障害（運動後の誘導の遅れ）と糖代謝異常について確認されたため，IL-1によるIL-6誘導能の運動負荷による影響についても検討した．結果，IL-1によるIL-6誘導は運動負荷後において上昇することが示された．また，運動負荷後の糖取り込みに加えて，筋組織内グリコーゲン量についても，IL-1β投与により上昇した．以上の結果から，筋運動時におけるIL-1の機能的役割について，（１）運動時に筋組織のMφにおいて発現・誘導され，（２）オートクライン/パラクラインに筋組織に作用することで，（３）筋細胞レベルでのエネルギー確保に重要な作用を有することが明らかとなった．
本研究結果は運動時のMφの筋機能維持における重要な役割を示す所見であり，軽度な筋組織外傷といえる咀嚼様運動に伴う筋疲労時に，疲労回復過程においてMφが必須な存在であることを示すものである．
