小胞体ストレスに着目した筋萎縮性側索硬化症の治療法開発

Outline of Annual Research Achievements

近年、Matrin-3（MATR3）遺伝子のミスセンス変異が声帯および咽頭麻痺を伴う遠位型ミオパチー (VCPDM) および遺伝性筋萎縮性側索硬化症（ALS）の原因として同定されたが、その発症メカニズムは不明である。我々は、MATR3変異を有するアジア初のVCPDM家系を経験した。本年度は、変異MATR3が骨格筋および運動神経障害を引き起こす病態を解明することを目的とした。方法として、１）初代筋芽細胞あるいはC2C12 細胞に野生型もしくは変異型 (S85C) MATR3、あるいはMATR3に対するsiRNAを導入し、細胞内局在および細胞増殖性、DNA損傷マーカーであるリン酸化ヒストン2AX (γH2AX)、リン酸化Trans-activating response region DNA-binding protein of 43 kDa (TDP-43) 発現を評価した。また本邦初のVCPDM 自験2症例の骨格筋組織を用いて、MATR3およびp62/SQSTM1、TDP-43、ユビキチン、γH2AXの発現を検討した。野生型MATR3 は核に局在する一方、変異型MATR3 は細胞質に局在し、変異型MATR3導入細胞の増殖性の低下を認めた。またsiRNAによるMATR3の発現抑制によりγH2AXの発現の増加や、TDP-43のリン酸化および細胞質への局在変化を引き起こした。VCPDM 患者骨格筋では、核でのMATR3発現が低下し、変性筋においてp62/SQSTM1、TDP-43、ユビキチンの筋線維内凝集を認めるとともに高頻度にγH2AXの発現が見られた。変異型 MATR3は細胞質に異所性局在を示す結果、核での機能損失によりDNA 損傷を引き起こす一方、細胞質での異常蛋白凝集やTDP-43のリン酸化や細胞質への局在変化により毒性を獲得する可能性が示された。現在、本患者よりiPS細胞を作成し、運動神経および骨格筋への分化実験を行い、病態モデルの確立を目指している。
