Outline of Annual Research Achievements

本課題では、ペリオスチンが炎症の進展過程で果たす機能を明らかにするため、ペリオスチンとその細胞表面受容体との結合様式を解明するとともに、ペリオスチンを標的とした阻害抗体を作製して新規アレルギー治療薬開発の基礎的研究を行うことを目的としている。本年度は以下の項目を実行した。
1. ペリオスチンとインテグリンαVβ3との結合様式の解明：NIH/3T3細胞にNF-κB－ルシフェラーゼレポーターを組み込んだ細胞を作製し、この細胞のペリオスチンをsiRNAでノックダウンしたところ、ルシフェラーゼ活性の低下が認められた。またこの細胞にペリオスチンを強制発現させるとルシフェラーゼ活性は上昇した。すなわち、ペリオスチンはNF-κB経路を活性化することが確認された。これがインテグリンαVβ3を介しているかどうかを、阻害剤を用いて検討中である。
2. インテグリンαVβ3以外のペリオスチン結合分子の同定：フローサイトメトリーでペリオスチンの細胞との結合を調べる系を構築し、インテグリンβ3が発現していなくてもペリオスチンと結合する細胞を見出した。この結合に関与する分子を同定中である。
3. ペリオスチンの阻害抗体の作製：継続中であるが、現在のところ目的の抗体は得られていない。
