炎症性歯周組織におけるＴｈ１７細胞に対するＩＬ－３５の影響

Outline of Annual Research Achievements

歯周病病態でのTh17関連免疫応答におけるIL-35の役割を明らかにすることを目的として、歯周炎患者および健常者サンプルを用いて検討を行った。
平成26年度は、平成24、25年度に引き続き、歯周炎患者および健常者の末梢血を採取し、そこからCD4+T細胞をソーティングし、Th17細胞誘導条件下で検討を行った。歯周炎患者あるいは健常ボランティアのCD4+T細胞より誘導したTh17細胞にrIL-35を添加し、RORa、RORc、IL-17AのmRNA発現を比較したところ、rIL-35添加によりRORa、IL-17AのmRNA発現の抑制傾向がみられた。この検討では市販のrIL-35に加え、我々の研究室で精製したrIL-35においても検討を行ったが、同様に抑制傾向を示す結果を得ている。また、同じ実験系において、IL-35の受容体であると考えられているgp130 (CD130)およびIL-12Rβ2のmRNA発現を検討したところ、Th17細胞への誘導およびrIL-35の添加による発現の変化は認めなかった。。
また、歯周炎患者および健常者の末梢血においてTh細胞ポピュレーションの比較検討を目的とし、末梢血からCD4+T細胞をソーティングし細胞内サイトカインFACSにより検討を行った。その結果、歯周炎患者では健常者に比較し、IL-17AおよびIFN-γ産生細胞が多く、Foxp3陽性細胞は少ないという傾向を得ている。しかしながら、まだサンプル数が少なく、今後サンプル数を増やした上で、解析を行う必要が有る。
