薬物刺激間葉系幹細胞を用いたＤｕａｌ Ｓｔｅｍ Ｃｅｌｌ Ｔｈｅｒａｐｙの開発

Research Abstract

間葉系幹細胞を応用した細胞治療は培養増幅した細胞を体内に投与する方法が一般的である。我々は培養増幅した間葉系幹細胞をテリパラチドで刺激することで、その造血支持能力が増強することを見いだした。ヒト間葉系幹細胞（mesenchymal stem cell: MSC）をヒトCD34陽性造血幹前駆細胞（hematopoietic stem/progenitor cell: HSPC）とSCF（stem cell factor）、TPO（thrombopoietin）、IL-3（interleukin-3）、Flt3-L（Flt3-ligand）存在下で共培養すると HSPCがin vitroで増幅する。MSCをテリパラチドで薬物刺激したのちに、同様にサイトカイン存在下でHSPCとの共培養を行うとHSPCの増幅が増強した。マイクロアレイ解析を行ったところ、テリパラチドで刺激されたMSCはカドヘリンファミリー分子の一つであるOB-カドヘリンの発現が増強していた。OB-カドヘリンに対するsiRNAおよび中和抗体を用いた機能実験で、HSPCの増幅増強が抑制されたこと、OB-カドヘリンはHSPCにも強発現していたことからOB-カドヘリンのホモフィリックな結合を介した MSCとHSPCのcell-cell interactionがHSPCの増幅増強に関与していると考えられた。以上から、テリパラチドによる薬物刺激がMSCの機能を増強することが示された。
