計算生体力学による微小循環系の細胞の機能と環境の相互作用の解明

Outline of Annual Research Achievements

微小循環系における細胞の流動および細胞の接着現象を計算するため,赤血球を始めとする細胞を内部流体と超弾性膜からなるカプセルで近似し,膜の固体力学に対する有限要素法と,細胞内外の流体力学に対する境界要素法あるいは格子ボルツマン法と連立し,さらにリガンド‐レセプタ結合をモンテカルロ法で計算する計算手法を開発した.全てのプロセスを Graphics Processing Unit(GPU)を用いて計算することで,大規模なパラメトリック計算を可能にした.
壁面近傍における細胞挙動について境界要素法に基づく解析を実施し,細胞のリフト速度に対するせん断速度勾配と壁面の寄与の関係を明らかにした.また振動流中における細胞の変形量を解析し,振動流中では定常流中に比べて過剰な変形を示すことを明らかにした.
血管内の細胞の力学環境を明らかにするため,単純せん断流れ場に対するカプセル懸濁液のレオロジー解析を実施した.大規模パラメトリック計算によって,せん断速度に対する見かけの粘度の変化を定量化した.また白血球や血小板などサイズや硬さの異なるカプセルが混合している懸濁液に対しても同様の大規模パラメトリック計算を実施し,見かけの粘度を定量化した.
白血球やがん細胞の微小循環における流動形態および流速を解析し,これらの細胞がMarginationした場合においても,血液の平均流速よりも高い速度で流動していることを明らかにした.またこれらの細胞と血管内皮細胞間のリガンド‐レセプタ結合をBellモデルに基づくモンテカルロ法で記述し,微小循環内の回転運動および定常接着を再現した.
