Outline of Annual Research Achievements

我々は、インスリン/インスリン様成長因子(IGF)の重要なシグナル伝達因子であるインスリン受容体基質(IRS)が、インスリン/IGF刺激非依存的に細胞内で多種類のIRS結合タンパク質(IRSAPと命名)と相互作用し、巨大な分子複合体(IRSomeと命名)を形成しており、他の因子の刺激や生体が置かれた生理状態に応答してIRSomeの構成タンパク質がダイナミックに変化し、インスリン/IGFシグナル・生理活性を変化させていることを発見した。これまでに我々は、IRS-1と相互作用する分子のスクリーニングにより、クラスリン依存性エンドサイトーシスのアダプター分子AP2を同定している。今回IRS-1とAP2の相互作用の意義を検討した。IRS-1を過剰発現するとIGF-I受容体(IGF-IR)のエンドサイトーシスが抑制され、この抑制は結合部位を変異させてAP2と結合できないようにしたIRS-1変異体の過剰発現では観察されなかった。逆にIRS-1を発現抑制すると、IGF-IRのエンドサイトーシスが亢進したことから、IRS-1がAP2との相互作用を介してIGF-IRのエンドサイトーシスを阻害することを見出した。他の結果も併せると、インスリン様シグナル経路下流のセリン/スレオニンキナーゼmTORC1依存的なIRS-1の分解によって、Aktなどの下流シグナルの抑制に加えて、IGF-IRのエンドサイトーシスも引き起こされることで、IGF活性の発現調節が可能になっていると結論した。本年度、DGKzとIRS-1との相互作用を抑制する低分子化合物の取得に成功し、これを用いた相互作用の生理的意義の解明を進めた。更に、アミノ酸欠乏培地で培養した肝細胞でインスリンが存在しない状態でもこれまでに報告のない機構でインスリン様活性が発現することを発見した。

Reason

IRSに相互作用する新しいIRSAPの同定に成功し、これらのインスリン様活性の調節に果たす役割の解明が順調に進んでいる。今回、IRS-1がAP-2と相互作用することにより、IGF-I受容体のエンドサイトーシスを抑制し、IGFシグナルが長期間持続する可能性を示すことができた。こIGFがなぜ長期作用を発揮するかの機構は全く明らかにされていないが、これを新しい観点から明らかにできた点は特記に値する。また、アミノ酸が欠乏状態で飼育あるいは培養された動物の肝臓や肝臓細胞では、インスリン様活性が観察されるが、この一部は、これまでのインスリン様シグナル伝達異なる経路で誘導されることも明らかにしており、期待以上の結果が得られている。

Strategy for Future Research Activity

今回、同定・選択したIRSAPについて、特に、DGKz、GKAP42とIRS-1の相互作用を修飾する低分子化合物の取得に成功したので、これらの化合物が脂肪細胞などのインスリン様活性の変動に及ぼす影響を調べる。
アミノ酸が欠乏状態で飼育あるいは培養された動物の肝臓や肝臓細胞では、インスリン様活性がどのような機構で誘導されるか、これにIRSやIRSAPは関係するのかも併せて検討する。
また、IRSと選択されたIRSAPの発現を過剰・抑制した細胞・動物、IRSと当該IRSAPの相互作用を修飾した細胞・動物におけるインスリン様シグナル・インスリン様活性の解析についても、これまでの方針どおり、研究を進める。
