Outline of Annual Research Achievements

全ゲノムシークエンス、全エキソーム、パネルを用いたがんの臨床シークエンス体制が完成し、本格運用をした。シークエンサーはPrizm 3730、Miseq、Ion PGM、Ion Proton、及びHiseq2500から構成される。ヒトゲノム解析センターのスパコン（2015年からはShirokane3）の一部を論理的に切り出し、インターネットから切り離された高セキュリティデータ解析専用領域（グリーンエリア）を作った。専用データ解析室及びコントロール室（外部とのデータ入出力を行う）を整備した。データ解析室のPCはグリーンエリアにつながっているが、USBなどの口がなく、データの入出力できないシンクライアントのものを整備した。サンプル調整からデータ解析結果まで一元管理し、解析のトレーサビリティ―を担保し、ドキュメンテーションを行うために、Genlogic社のClarity LIMSを導入して改良した。アクセスは、二重の生体認証システムと専用ICカードにより三重のセキュリティとなっている。データベース検索などのための外部との接続にはノート型PCを持ち込むが、専用のWiFiコントロール下に置いている。グリーンエリアから臨床系のオフィスへはVPN接続をして利便性を図るが、オフィスへのアクセス及び専用シンクライアントPCへのアクセスには二重の生体認証システムをかけた。このグリーンエリアに、サンプル調整、シークエンス、データ解析等の機能を全て入れることとした。研究用としてIBMワトソンを北米を除いて世界で初めて導入した。この体制で、情報、血液腫瘍、消化器がん等の固形がん、ELSI、スパコンSEからなる約25名のチームが臨床シークエンス研究を実施した。その結果、医師による精密診断の高精度支援の実現、がん研究における新たな発見があり、今後、論文、人類遺伝学会、血液腫瘍学会等で今後発表する。
