Outline of Annual Research Achievements

本研究は、脳に発現する血小板由来増殖因子（PDGF）が、脳組織の再生や修復を誘導する機序を解明する。同受容体（PDGFR）の条件的ノックアウトマウス（KO）を用い、１）PDGFR‐bが側脳室下帯（SVZ）由来の新生神経細胞の脳梗塞病変への動員、２）PDGFR-aが、稀突起膠細胞の前駆細胞（OPC）がSVZより分化、増殖し髄鞘形成細胞に成熟し、脱髄疾患を修復する過程で果たす役割を、それぞれに明らかにすることを計画した。
１）ではNestin Promoterにより誘導されるCre recombinaseが誘導する神経細胞特異的なPDGFR-b KOマウスの脳梗塞巣に、これまでに報告のない高度の神経新生が見られることを見出した。この高度の神経新生は神経幹細胞におけるIntegrin発現の亢進の結果として見られる細胞遊走能の亢進と、脳梗塞巣における神経幹細胞の増殖因子であるCXCL12発現の亢進という要因が複合してもたらされたことを示唆するDataとともに論文として報告した（Sato et al., Stem cells, in press）。梗塞脳にはこれまでの予想を上回る再生能力があることを示したもので、社会的な反響も大きく、患者様からの問い合わせも頂いた。Mediaからの反応も複数あり、読売新聞では全国版に報道されている。
２）ではGenetic mapping, Virus vectorを用いたLineage tracing等の手法により髄膜や脳実質の血管周囲に分布する間葉系幹細胞由来の未熟な細胞がPDGFR-a 依存性に脳に動員されてOPCの欠失を補うことを示した。Adult brainでOPCの全Populationが一度に置き換わる規模の再生能力であり、髄鞘が傷害される疾病の治療に道を開くものである。論文の投稿中で、受理に向けて努力をしているところである。

Reason

二種類のPDGF受容体がそれぞれ独立して、これまでに報告がない新しい様式の脳神経組織再生に寄与する経路を見出しており最終年度での完成を目指している。

Strategy for Future Research Activity

我々が提示している間葉系幹細胞の脳実質への動員と稀突起膠細胞の動員という経路は全く新しい細胞動態であり、雑誌への受理に困難をきたしている。提示している仮説をより強いものにしていくためにより詳細な解析を加えて論文の受理への努力を継続したい。

見出した再生能力を疾病の克服に向けて応用していくことが今後の研究の課題である。
