Outline of Annual Research Achievements

今年度は昨年度に引き続き線維芽細胞におけるペリオスチンによるNF-kB活性化機構について解析を進めている。NF-kBプロモーター領域配列を含んだレポーター遺伝子の強制発現細胞株を用いてペリオスチンによるNF-kB活性化機構について解析を行っている。その結果、ペリオスチンを強制発現させるとNF-kB活性化が亢進し、ペリオスチンをノックダウンするとNF-kB活性化が低下し、NF-kB活性化の程度がペリオスチンの発現量と比例していることが明らかとなった。今後は、ペリオスチン受容体の同定、NF-kB活性化に至るシグナル伝達分子の同定を進める予定である。
また、この強制発現細胞株を用いて阻害剤の開発も進めている。阻害抗体の探索を行うとともに、製薬企業との共同研究により、低分子化合物の効果について検討を進めている。さらに、RNAiの手法を用いてがペリオスチントランスジェニックマウスにおいてペリオスチン発現が低下することを見出し、今後モデルマウスでの解析を進めて行く予定である。

Reason

ペリオスチンによるNF-kB活性化機序を解析できるin vitroの実験系を確立しており、今後解析を進めることで、その活性化経路を解明できると期待される。
ペリオスチンに対する阻害剤において、阻害抗体については引き続き作製中であると、低分子化合物については候補物質を獲得しており、今後モデルマウスを用いて解析を可能としている。さらにRNAiがトランスジェニックマウスにおいて有効であることも見出しており、やはり同様に今後モデルマウスを用いて解析を可能としている。

Strategy for Future Research Activity

ペリオスチンによるNF-kB活性化機構についてレポーター遺伝子の強制発現細胞株を用いて解析を進める予定である。
低分子化合物、RNAiをモデルマウスに適用してその効果を検討するとともに、引き続き阻害抗体の探索を進める。
