OH-PCB妊娠期曝露による体内代謝経路と次世代への甲状腺ホルモン攪乱作用の解明

Outline of Annual Research Achievements |
妊娠中・胎児期の環境要因が児の成長発達や疾病発症に及ぼす影響を解明するために立ち上げた一産院における前向き出生コホート(N=514)を用いて、OH-PCBの母体血中濃度測定と代謝関連遺伝子多型(SNPs)の解析と評価を行った。SNPs(AhR、CYP1A1、CYP1B1、GSTM、GSTT)で層別化した集団間での4-OH-CB 146 + 3-OH-CB 153、4-OH-CB 187およびΣOH-PCB母体血中濃度差を検討したところ、遺伝子多型による有意な濃度差はみられなかった。 妊婦514名のコホート参加者のうち、妊娠中母体血中OH-PCB濃度と妊娠中の母親および出生時の札幌市による甲状腺マススクリーニング測定結果から得られた甲状腺ホルモン値データ(TSH、FT4)が揃う260組の母児において、OH-PCBの体内濃度が母児甲状腺ホルモン値へ与える影響について解析・検討を行った。独立件数は、母体血中∑OH-PCB、4-OH-CB 146 + 3-OH-CB 153、4OH-CB187濃度、従属変数は、母児TSH、FT4値とし、共変量に母親の出産時年齢、非妊娠時BMI、出産回数、魚摂取量、世帯年収、OH-PCB測定用採血時期と、児の解析のみ在胎週数、出生時体重、生下時採血日数を加えて重回帰分析を行った。その結果、母において母体血中4-OH-CB187濃度(log10)が高いほど、FT4(log10)が高く(β=0.033、p=0.044)、児においてはΣOH-PCB、4-OH-CB187濃度(log10)が高いほど、FT4(log10)が高かった(β=0.031、p=0.046、β=0.029、p=0.017)。児の男女別解析では、男児でΣOH-PCB、4-OH-CB187濃度(log10)が高いほど、FT4(log10)が高かった(β=0.053、p=0.017、β=0.036、p=0.043)。女児では有意差が認められなかった。 コホート参加者のうち、臍帯血検体を有する264名について、臍帯血中レプチン・アディポネクチン濃度の測定を行った。総アディポネクチン濃度の平均値は19.2μg/ml、高分子量アディポネクチン濃度の平均値は12.8μg/ml、レプチン濃度の平均値は7.42μg/mlであった。
