EGFR阻害剤の副作用を低減する頭頚部扁平上皮癌併用療法の開発

Outline of Annual Research Achievements

平成26年度は脳血管攣縮治療薬ファスジルと分子標的薬セツキシマブの併用における頭頚部扁平上皮癌細胞（HNSCC）への影響について解析を行った。
HNSCC株であるHSC-3にセツキシマブの添加を行い24時間後にファスジルを添加し，さらに24時間後のCXCL14遺伝子発現量及びタンパク分泌量を解析した。その結果，セツキシマブの添加群で遺伝子発現が添加濃度依存的に亢進し，ファスジルの添加群でBRAKの細胞外分泌量・分泌された割合が添加濃度依存的に亢進した。共添加群においてはその両方が亢進していることが確認された。
セツキシマブとファスジル共添加における細胞毒性などを検討するため，HSC-3にセツキシマブの添加を行い24時間後にファスジルを添加し，さらに24時間後の細胞数の測定を行った。その結果，control群，セツキシマブ添加群，ファスジル添加群，共添加群において細胞数の有意な差は認められなかった。
これらの結果より、セツキシマブとファスジルの併用は、血管新生抑制因子であるCXCL14のタンパク合成亢進，分泌促進の両方に作用し新たな多剤併用療法として応用できる可能性を示す知見である。
