地球上の全ての生物の進化は、飢餓や低栄養に対する適応の歴史であり、生物の寿命はこれらの環境因子によって規定されてきた。実際、低栄養やそれに伴う免疫力の低下、感染症がヒトの死亡の主因を占めていた。ところが20世紀後半より、歴史上経験のない過栄養の時代を迎え、その過栄養が生活習慣病などの疾患を引き起こし、寿命短縮の原因になっている。このような短期間の劇的な栄養・環境の変化によってもたらされる問題の抜本的解決には、様々な栄養・環境状態における生物の普遍的生命現象の根源の俯瞰的解明と、その破綻メカニズムの理解が不可欠である。
本研究課題では、健康長寿のための普遍的代謝経路の解明とその実現への方法論を確立することを目的とする。
1、寿命延長効果をもたらす高等生物に適したカロリー制限の科学と方法(栄養素の量と質)を明らかにする。 : 摂取カロリーの最適な「質」を決定するため、カロリー制限を一定とし、炭水化物、タンパク質、脂質の割合を変動させ、それぞれのマウスにおける寿命の検討を継続した。様々な栄養条件下での寿命関連臓器や代謝関連臓器における生物学的反応やその代謝調節機構において、栄養シグナルの同定を開始した。
2、健康長寿を制御する普遍的シグナルを同定する。 : AMPK/SIRT1依存的/非依存的寿命延長経路のそれぞれの細胞内シグナル、臓器間ネットワークをAdipoR活性化低分子化合物とAdipoR各種遺伝子改変動物(全身/組織特異的AdipoR欠損/過剰発現マウス)を組み合わせ、メタボローム・トランスクリプトーム解析などを行い、個体レベルでの代謝パラメータなどを解析し、寿命延長効果の有無を検証した。
3、代謝制御経路を基盤にした健康長寿実現に向けた科学と方略を確立する。 : AdipoRの立体構造解析に成功し(Nature, 520, 312, 2015)、これらの情報を利用し、構造機能相関解析を進めた。

申請時、予定していた研究項目((1) 寿命延長効果をもたらす高等生物に適したカロリー制限の科学と方法(栄養素の量と質)を明らかにする。(2) 健康長寿を制御する普遍的シグナルを同定する。(3) 代謝制御経路を基盤にした健康長寿実現に向けた科学と方略を確立する。)のうち、(1)、(2)については、それぞれ平成27年度に予定していた計画内容について、当初の計画通り進展している。(3)については、平成27年度に予定していた計画内容以上に研究の進展があり、成果として、Nature誌に論文を掲載した。

平成27年度については、当初予定していた計画内容について、ほぼ達成した上、さらに一部の研究計画については、当初の計画以上に進展している。そのため、本研究の推進において、研究体制及び研究計画に大幅な変更の必要性は低いと思われ、当初の計画通り、研究を推進する予定である。本研究課題のさらなる進展・加速を目指すべく、研究分担者・研究連携者を含めた各人の実験の精度の向上とスピードアップを図り、目的を早期に達成したいと考えている。
