Outline of Annual Research Achievements

本研究は新しい化合物-標的相互作用検出技術を開発し、未解明の天然物の標的分子と作用機構を明らかにすることを目的とする。
（１）化合物の標的分子を同定するために「非必須」遺伝子破壊株だけでなく、「必須」遺伝子の条件致死遺伝子変異株ライブラリーや二倍体ヘテロ型遺伝子破壊株ライブラリー、遺伝子過剰発現ライブラリーの増殖をバーコードシークエンスにより定量することで、より精度よく標的分子を同定できるよう改良した。
（２）Pooled shRNAウイルスライブラリーを用いた標的分子の高速スクリーニング技術について、シークエンスライブラリー調製時のPCR産物精製法の改良を行ったところ、マルチプレックスバーコードシーケンス法におけるバーコードリード数が上昇して多検体同時シークエンスが可能になった。この系を用いてユニークな天然生理活性物質の作用機序解析、既知化合物との合成致死遺伝子のスクリーニングに取り組んだ。
（３）SIRT2は脱アセチル化酵素活性だけでなく、脱長鎖アシル化酵素活性を持つが、一旦脱長鎖アシル化反応が起こると、脱アセチル化活性を失うことがわかった。X線結晶構造解析の結果、脱長鎖アシル化反応の生成物であるO-アシル-ADPリボースが酵素ポケットに強く結合し、脱アセチル化阻害剤となることが明らかになった。一方、基質がアシル化ペプチドの場合には、O-アシル-ADPリボースをキックアウトするために反応が進むことがわかった。すなわち、基質が生成物の解離を促進する新たな反応機構であることが示された。
（４）NanoBiTを用いた細胞内化合物―標的同定法をさらに強化するため、新たに約2,000種のヒトcDNAを追加し、合計12,000クローンでのスクリーニングが可能となった。
（５）ケミカルエピジェネティクス研究用化合物の安定供給のため、生合成遺伝子クラスターを利用した異種発現系を確立した。
