Elucidation of pathogenic immune memory formation and maintenance

Outline of Annual Research Achievements

IL-33によって発現誘導される新規機能分子であるAmphiregulinに着目し、生体内におけるAmphiregulinによる病原性誘導機構の解析を行った。その結果、
Amphiregulinを特異的に産生し、気道の線維化に深く関与する記憶型Th2細胞を同定し「線維化誘導-病原性記憶CD4 T細胞」と命名し論文発表を行った(Morimoto et al. Immunity 2018)。その後、プレスリリースで社会への発信を行い、NHKなどで報道され注目を浴びた。研究成果の概要としては、同細胞から産生されるAmphiregulinは、好酸球に発現するEGF受容体を介して、好酸球を活性化し、炎症性好酸球へリプログラムする。この結果、好酸球は、細胞外マトリックスの構成成分として重要なOsteopontinを産生し気道周囲の線維化を誘導する。これら、IL-33, Amphiregulin, Osteopontinのどの分子も好酸球性気道炎症における気道周囲の線維化に必須であることを明らかにした。さらに、同様のAmphiregulin産生記憶型CD4 T細胞およびOteopontin産生好酸球が、好酸球性副鼻腔炎患者由来のポリープ中にも多数認められることを明らかにした。以上のことからヒトの慢性好酸球性炎症疾患における組織線維化の病態形成においても“IL-33-Amphiregulin-EGFR-Osteopontin axis”が深く寄与している可能性が示唆された。
さらに、ポリコーム及びトライソラックス分子群による記憶Th細胞の機能変換・維持機構に関する解析を行った。ヒストンのリジン4トリメチル化(H3K4me3)活性を有するトライソラックス複合体の中でもCOMPASS複合体の一員であるCxxc1タンパク質に着目し解析を進めている。論文発表を近々行うべく準備中である。
