Outline of Annual Research Achievements

機能的メモリーB細胞の前駆細胞は胚中心(GC)B細胞であるが、本年度は胚中心B細胞で顕著な発現上昇が認められる転写因子Foxo1のGC B細胞における役割、メモリーB細胞への分化における役割を検討し、以下の成果を得た。1)Foxo1遺伝子のpre-GC, GC phase特異的な欠損をおこなうと、pre-GCではFoxo1遺伝子欠損さすことにより、抗原特異的B細胞の、より過剰な増殖が見られた。逆にGC phaseではGC細胞の増殖phaseであるDZ GC B細胞が観察されず、又, それによりGC B細胞全体の増殖が低下した。2)メモリーB細胞への分化は、Foxo1欠損により、あまり変化は認められなかった。3)GC B細胞の増殖不全は、十分量のT細胞ヘルプを供給することによっても生じ、GC B細胞内でのシグナル異常が原因で、DZ GC B細胞が生成されないことが判明した。4)T細胞ヘルプによりGC B細胞が増殖するためには、先ず増殖initiator転写因子の発現上昇が必要と考えられているが、c-Myc, BATFの転写因子共に、Foxo1依存性に発現が上昇し、又、これら２つの転写因子の発現上昇はGC B細胞の増殖に必須であることが判明した。以上の結果は、PI3kinase活性化によりFoxo1遺伝子活性が減弱するわけであるが、pre-GCにおいてはPI3kinaseの活性の強度に依存して、B細胞増殖が上昇するが、GC細胞では必ずしもそうではなく、「GC反応中においては、適切な強度のPI3kinaseシグナルが、GC B細胞の増殖に必要である」ことを強く示している。

Reason

予測以上に早く、世界に先駆けて、新規概念の提出(メモリーB細胞生成メカニズム; Nature Immunology (in press))にまで至った。しかしながら、メモリーリスポンスを支える細胞・分子メカニズムの解析は、マウスの樹立がやや遅れ気味である。しかし、このマウスもできつつある。全体的には順調に進展している。

Strategy for Future Research Activity

今までの、やや遅れている面は（メモリーリスポンス）、この解析技術には十分習熟しているので、一気に進め遅れを挽回するよう努める。又、予測以上に先行しているパート(メモリーB細胞生成メカニズム)は、更にその次の重要な課題も明確になっていると同時に、必要な実験材料も樹立しているので、常に国際的にリードを保つべく進展さす。
