| Outline of Annual Research Achievements |
1.オートファジー選択的基質p62/Sqstm1がユビキチンリガーゼアダプタータンパク質Keap1を不活性化し、Keap1のターゲットであるストレス応答性転写因子Nrf2を活性化することを報告してきた。このp62/Sqstm1-Keap1-Nrf2経路が肝細胞がんにおいて活性化していること、この活性化がグルクロン酸経路およびグルタチオン合成を亢進させ腫瘍の増殖、抗がん剤耐性に寄与することを見出した。また、p62/Sqstm1-Keap1-Nrf2経路を標的にした抗がん剤スクリーニングを実施し、ヒット化合物を得た。これらの結果をまとめ論文として発表した。
2.上記のヒット化合物の誘導体の作成、細胞内評価を行った。
3.肝臓特異的オートファジー欠損マウスを用いたリピドーム解析から、オートファジー欠損肝臓において脂肪酸酸化の障害が起こっていることが判明した。これは新規オートファジー選択的基質の蓄積が原因であることを見出した。
