Critical role of newly identified S100A8/A9 receptors in lung specific cancer metastasis

Outline of Annual Research Achievements

・Soil signal(S100A8/A9)-Soil sensor(SSSRs)の意義を、動物モデルにおいて確認する
S100A8/A9が働く受容体はTLR4とRAGEのみではないことが判明し、S100A8/A9 タンパク質に対する特異的受容体の新規同定を試みた。その結果、EMMPRINの発見に加え、4つの新規受容体「NPTNα,NPTNβ, MCAM, ALCAM」も同定することができた。これらの受容体群を新規S100 soil sensor receptors (SSSRs)と名付けた。当新規SSSRsは、種々多様ながん細胞で発現が上昇している特性を示すが、これら受容体が共通して発現しているメラノーマの細胞株を用いて尾静脈投与によるマウス肺転移実験を行った。その結果、EMMPRIN、NPTNβ、MCAMを移植がん細胞に過剰発現させることにより、肺転移性が顕著に増加し、逆にこれらのドミナントネガティブ体（機能阻害）の過剰発現により肺転移性が顕著に減少した。当動物実験より、EMMPRIN、NPTNβ、MCAMは、その発現が上昇しているがん種においては、治療の有効な分子標的となりうる可能性があることが示された。

・Soil signal(S100A8/A9)-Soil sensor(SSSRs)の連携をブロックする手法を構築する
試行錯誤から、S100A8/A9を吸着し、がん細胞に作用させないデコイタンパク質製剤{受容体細胞外領域(exReceptors)とIgGのFc領域との融合キメラタンパク質}がブロック製剤として有効となる可能性を見出した。独自に作成したexEMMPRIN-Fc, exNPTNβ-Fc, exMCAM-Fcは、メラノーマの走化、浸潤能の抑制に優れていることを確認した（In vitroの解析）。
