| Outline of Annual Research Achievements |
ATL細胞におけるエピゲノム異常(ヒストンのメチル化、ユビキチン化、アセチル化、RNApol IIの結合パターン)、NF-kBなどの転写因子群の結合パターン、及びATL及びHTLV-1感染細胞のtranscriptomeを複合的に解析し、ATL細胞における遺伝子発現パターン及びその原因を明らかにした。さらに転写制御の起点となるEZH1/2の異常を同定し、さらに新規EZH1/2阻害剤の開発と臨床研究への橋渡しにも成功した。
またATL細胞におけるTAK1-p38-eIF4Eの活性化、eIF4Aの活性化、及びmicroRNAの大規模な発現減少をそれぞれ発見し、ATL細胞における遺伝子翻訳の異常な活性化の存在を明らかにした。さらにリボソーム解析と発現アレイを組み合わせた翻訳標的遺伝子群の同定に成功し、ATL細胞の新たな特徴と分子標的としての可能性を明らかにした。
ATL細胞のmRNAスプライシング異常について、健常人CD4+ T細胞(n=8)と急性型ATL患者PBMC (n=16)でエクソン・アレイ解析を行い、ATL細胞での約10,000 mRNAのエクソン欠損・重複を見出した。次にATLで過剰発現が顕著なmRNA (CADM1, EVC, MAP3K14, MYB, TIAM2など)にエクソンが欠損するパターンが集積していることを見出し、異常タンパク質の過剰発現の可能性を示した。また我々の遺伝子発現アレイ解析（GSE33615）との統合解析からATL細胞では多くのスプライシング・ファクターが発現低下しており、スプライシング制御不全との関連が示唆された。さらにHAS-Flow法によりATL患者より分取した細胞でもエクソン・アレイ解析を行い、不死化・腫瘍化過程でのスプライシング異常の蓄積の様子を明らかにしつつある。
以上、本研究によりHTLV-1感染細胞の多段階発がん過程における遺伝子制御異常の実態が明らかになった。
