Outline of Annual Research Achievements

アフリカ地域において、感染症制圧は依然として緊急性の高い課題であり、我が国からも精力的にその研究・支援が行われている。2006年、WHOとUSAIDは病原体媒介生物のコントロールのため、有機塩素系農薬であるDDTの屋内限定散布(IRS)を推奨した。即ち、現在のVector Controlにおいて、DDTに代替えする薬剤は限られており、その年間散布量は、アジア・アフリカ地域を中心に5,000トンに達していると報告されている。その結果、マラリアによる死亡率は、2000年に比べ、世界で25％、アフリカでは33％減少し、一定のbenefitが得られたことが報告された。その一方で、IRSは屋内に薬剤を直接散布するため、そのヒト健康影響が懸念されているが、その影響は不明な点が多い。更に、IRSではDDTのみでなく同時に様々なピレスロイド系農薬も散布するが、その共暴露に伴うヒト健康や、希少野生動物が生息するアフリカの生態系に対するriskは不明である。すなわち、DDT・IRSについて、より詳細なrisk-benefit解析を散布が推奨されてから7年が経過した今こそ行う必要がある。
以上の背景の基、本研究の目的は、「Vector control地域におけるDDTやピレスロイドの中・長期使用時のヒト健康および生態系へのriskを予測する」ことである。
2014年度は、マラリアコントロール地域であり、実際にDDT・IRSが行われている南アフリカのクワズルナタール・ンドゥモで、カウンターパートであるヨハネスブルグ大学およびノースウェスト大学と共にサーベイランスを実施した。本調査では、野生動物（魚類、両生類、鳥類、げっ歯類）と家畜・家禽に注目し、試料採集を実施した。調査の結果、Topプレデター種に加え、ニワトリから高濃度のDDTが検出された。検出された濃度は、ヒトが摂取することで、発がんリスクが上昇するレベルに達していた。

Reason

2014年度は予定通り南アフリカ共和国・クワズルナタールで調査を実施し、魚類や両生類、鳥類、げっ歯類等の野生動物、ニワトリやウシ、ヤギ等のヒトの食料源となる家畜・家禽の採集を計画通り行った。更に、DDT散布地域を流れるポンゴラ川とその河口であるマプト湾での海産魚類の試料採集も実施した。得られた試料の日本への空輸も完了しており、既にDDTやピレスロイド農薬の化学分析を開始している。
また、今回の調査で特に、高濃度のDDT蓄積が見られた鳥類について、化学分析と共に、トランスクリプトームを中心としたバイオマーカー解析も開始しており、どのような毒性影響が出ているのか、遺伝子レベルで解析を行っている。
一方、当該研究では、アフリカ地域に生息する希少野生動物の中で、特に化学物質感受性が高い生物種を同定することも目標の一つに設定している。しかし、とりわけ大型の野生哺乳動物では、in vivoでの評価が困難であるため、当該研究では皮膚線維芽細胞を用いた新たな評価法の開発を行った。今回は、検査捕獲時に、ライオンやハイエナ等の大型食肉目の皮膚片を採取し、その初代培養を開始した。皮膚は、比較的低侵襲で採取できる組織であり、in vivoでの暴露実験が出来ない希少野生動物の化学物質感受性を評価するうえで最適と考えられる。培養した皮膚線維芽細胞の遺伝子発現プロファイルは、肝臓とは異なるものの、異物代謝第I相反応および第II相反応の発現は維持されており、本手法により、種間の相対的な化学物質感受性が評価できると考えられた。

Strategy for Future Research Activity

2015年度以降は、2014年度に引き続き、試料採集を実施する。環境試料（水・土壌・大気）、野生動物（無脊椎動物・魚類・両生類・鳥類・哺乳類）、産業動物（ウシ・ヤギ・ニワトリ）およびヒトの血漿、尿、また野菜や穀物などの食料品の採集を行う。特に、希少動物種として、各種両生類、クロコダイル等の爬虫類に加え、レパードやハイエナ等の食肉目の哺乳類、またハゲワシ、アフリカハゲコウ、ペリカン等の鳥類の捕獲を計画している。これらの動物種は、それぞれの生息環境で食物連鎖上のKey speciesであり、DDTが高濃度に蓄積している可能性が考えられるからである。更に、散布地域の住民について、その食生活や生活習慣を明らかにするため、アンケート調査を実施する。一部、先行的に小規模の調査行った結果、タンパク源の依存率として、鶏肉や魚が主であることが明らかになっており、これら試料についての詳細な検討が必要である事が示唆されている。
得られた試料は、日本に空輸後、先述した化学分析とバイオマーカー解析を実施し、必要に応じて、確立したin vitro化学物質感受性試験を行い、化学物質に対してハイリスク動物種（高蓄積、高感受性種）の推定を行う。
なお、希少野生動物のin vitor感受性評価については、皮膚線維芽細胞をiPS技術を用い、iHep（誘導性肝臓様細胞）に誘導することを試みる。iHepに誘導することで、異物代謝能をより肝臓に近い条件で評価することが可能になると考えられる。
