Outline of Annual Research Achievements

アフリカ地域において、感染症制圧は依然として緊急性の高い課題であり、我が国からも精力的にその研究・支援が行われている。2006年、WHOとUSAIDは病原体媒介生物のコントロールのため、有機塩素系農薬であるDDTの屋内限定散布（IRS）を推奨した。即ち、現在のVector Controlにおいて、DDTに代替えする薬剤は限られており、その年間散布量は、アジア・アフリカ地域を中心に5,000トンに達していると報告されている。その結果、マラリアによる死亡率は、2000年に比べ、世界で25％、アフリカでは33％減少し、一定のbenefitが得られた事が報告された。その一方で、IRSは屋内に薬剤を直接散布するため、そのヒト健康影響が懸念されているが、その影響は不明な点が多い。そこで、本研究では、「Vector control地域におけるDDTやピレスロイドの中・長期使用時の人健康および生態系へのriskを予測する」ことを目的とした。
2015年度は、2014年度に引き続きマラリアコントロール地域である南アフリカ共和国のクワズルナタール・ンドゥモで特に野生げっ歯類に注目し試料採集を実施した。野生げっ歯類はヒトと生活圏を共有しているため、得られた蓄積や毒性影響に関するデータは、DDT・IRSのヒトへの影響を外挿するうえで有益であると考えられる。
先ず、DDT濃度を分析した結果、肝臓中濃度で2700±2300 ug/kg wet weightと極めて高濃度の蓄積が観察された。また、その蓄積特性も特徴的であり、中間代謝産物であるDDDが主要異性体であった。本蓄積濃度は、ラットにおいて次世代への白血病や悪性腫瘍のリスクが懸念されるレベルであり、同生活圏に生活するヒトに対する毒性影響が懸念された。

Reason

南アフリカ共和国における調査研究は極めて順調に進行しており、これまで報告した野外調査に加え、生態影響評価を行ううえで重要となる現地生物を用いた毒性試験にも着手し始めている。また、2014年度に極めて高濃度のDDTの蓄積が観察された家禽について、in vivoレベルの暴露実験を実施し、マイクロアレイによりトランスクリプトーム解析を行う事でDDTに最適なバイオマーカーの探索を実施している。更に、毒性影響評価として包括的な生体ネットワークを解析するシステムバイオロジー解析も並行して実施している。
一方、エチオピアにおける調査は試料採集を含め、一部遅れており、今後の課題である。

Strategy for Future Research Activity

2016年度は、特に魚類を中心とした現地生物の実験動物化を行うと共に、その暴露試験を実施する予定である。また、2015年度に引き続き、環境試料（水・土壌・大気）、野生動物（無脊椎度物・魚類・両生類・鳥類・哺乳類）、産業動物（ウシ・ヤギ・ニワトリ）の採集を行う。一方、保護区で、希少動物種であるペリカンの卵は30サンプル程度採取済みであり、そのDDT蓄積レベルと共に、卵殻菲薄化との関係を明らかにする。
更に、2015年度に実施したトランスクリプトームの結果を詳細に解析し、DDT暴露に対する高感度バイオマーカーの探索を実施する。
