Outline of Annual Research Achievements

α-グルコシダーゼ阻害活性を示すトレスタチンおよびアミロスタチンは、それぞれ土壌放線菌Streptomyces dimorphogenesisおよびS. conglobatusから産生されるC7Nアミノサイクリトール化合物である。この類縁化合物であるアカルボースは、α-グルコシダーゼ阻害活性を示し、現在、2型糖尿病治療薬として上市されている。このことから、トレスタチンおよびアミロスタチンの構造を化学的に変換できれば、新たな糖尿病薬ができると期待されている。
トレスタチンおよびアミロスタチン生産菌の染色体DNAから遺伝子ライブラリーを構築した。また、ゲノムシーケンサーを用いてドラフト配列を取得し、両化合物の生合成に関わる遺伝子クラスターを特定した。さらに、この配列を元に作製してPCRスクリーニングを行うことにより、遺伝子ライブラリから目的のクローンを獲得することに成功した。
これら生合成遺伝子を解析したところ、アミノサイクリトール部分（C7N部分）を生合成する2-epi-5-epi-valiolone合成酵素や糖部分を繋ぐグルコシルトランスフェラーゼなどが含まれていることを明らかにし、生合成遺伝子クラスターのクローニングに成功した。現在、これら酵素群生合成に関わることを目的に、遺伝子破壊株の作製行った。また、これら破壊株や改変株を利用することにより新たなアミノサイクリトール類縁体の創生に挑戦している。
