脂肪酸シグナルを標的とする癌幹細胞ターゲティングの検討

Outline of Annual Research Achievements

膜受容体の同定されていないエライジン酸（トランス脂肪酸）の既知の長鎖脂肪酸受容体であるGPR40およびGPR120との結合性を検討すると両受容体ともエライジン酸によりインターナライズした。エライジン酸はコレステロール・ラフト内に多く移行するが、ラフト内にはGPR40/120とともにEGFRが存在し、エライジン酸処理によりリン酸化レベルが亢進した。GPR40/120からEGFRへのシグナル伝達にc-SRCリン酸化が関与していた。この結果、GPR40/120→c-src→EGFR→ERK1/2へのシグナル伝達が確認された。
エライジン酸が大腸癌細胞の転移促進作用を有することから癌幹細胞への作用が考えられた。そこで、幹細胞関連因子の発現を検討すると、EGFRの下流で、APC wild typeのCT26大腸癌細胞では、nucleaistemin発現亢進とEMT誘導が見られ、canonical Wnt signal活性化が認められた。一方、APC-nullのHT29細胞では、nucleaistemin発現亢進とnon-canonicalなWn5a, CD44発現亢進が認められた。このように、エライジン酸はGPR40/120を受容体としEGFRをトランス活性化し、癌幹細胞性を促進すると考えられた。

当初の研究計画に沿ってほぼ予定した進捗が得られていいる。脂肪酸イメージングのための標識については初回標識がシグナルが低く、再標識を施行しつつある。

Strategy for Future Research Activity

当初の研究計画に沿った平成28年度の計画、および、これまでの結果でより詳細な検討が望ましいものについて検討を行う。検討課題として、以下のものを計画している。
１）脂肪酸特異的miRNAの特定と標識遺伝子の同定、２）脂肪酸標識によるイメージングと細胞形態変化と転移能変化の評価、３）間葉系幹細胞への脂肪酸の作用と阻害による転移への効果、４）病理組織標本で検討可能な脂肪酸シグナチャーの抽出とヒト大腸癌組織標本による検討

Causes of Carryover

脂肪酸標識の再試行と解析に必要な試薬類の納品時期が平成28年度にずれ込む可能性があったため、敢えて平成27年度中に発注しなかった。
