Outline of Annual Research Achievements

癌構成における癌細胞と癌微小環境構成細胞の役割について明らかにすることが本研究の目標であった。昨年に続き転移性肝癌において癌細胞と癌関連線維芽細胞の相互作用を癌細胞・線維芽細胞をシングルレベルで画像化し、特に癌細胞が間質細胞に近接存在する状態下で癌細胞分裂像が認められることを視覚的に証明し、さらに転移性肝癌において非癌部よりも癌部および癌部辺縁にコラーゲン・線維性タンパクが多いことを報告した。微小環境の細胞の中でも特に癌関連線維芽細胞が癌形成・転移時に重要であることが証明され、癌形成時に重要な細胞であることが解明された。さまざまな癌種の癌転移時の微小環境を検討したところ、転移部位によって癌微小環境の細胞構成が異なること、また癌腫により微小環境を構成する細胞が異なることが明らかになり、すべての転移部位において癌関連繊維芽細胞が重要であるとは限らないことが解明され論文報告した。さらに癌細胞と癌関連線維芽細胞の細胞融合している現象も突き詰め、特に腹水中にはその融合細胞の割合が多く、何らかの役割が有ると予想され引き続き検討中である。　転移の際に癌エクソソームが転移巣を形成するときに、転移前に特に繊維芽細胞内に入り込み、転移する環境を形成することを視覚的に証明した。癌細胞株による相違と分離した癌細胞株よりエクソソームのみを分離し、miRNA の解析は未だ検討中である。さらに脂肪肝・非アルコール性脂肪肝などが増加傾向にあるため、背景肝の違いによる癌形成時および転移性癌の癌細胞および癌形成周囲細胞の相互作用を解明し、現在論文化を進めている状況にある。
