Analysis of Notch signal pathway in epidermal regeneration and its application to therapy

Outline of Annual Research Achievements

潰瘍が修復する過程で表皮細胞は形態が変化し、潰瘍表面を遊走して欠損部を被覆していく。私はこの過程でシグナル伝達分子Notchの発現が低下することを見出した。Notchは様々な細胞において、増殖や分化、生存を制御しているので、皮膚潰瘍の再生表皮におけるNotchの役割の解析を図った。
まずは、in vitroでの解析を進めた。Notchを過剰発現する一方、siRNAやinhibitorを用いてNotchの働きを抑制した。その結果、Notchの発現が低下しても、表皮細胞の増殖には大きな影響を及ぼさないことがわかった。一方、表皮の分化に伴って発現が誘導されるケラチン 1やケラチン 10の発現は低下することから、Notchの発現低下は表皮細胞の分化を抑制することがわかった。さらに、その発現低下によって、IL-36が誘導されることを見出した。
次に、in vivoで解析するため、マウス皮膚にNotchシグナルのinhibitorを塗布した。その結果、in vitroの結果と合致して、Notchを抑制することで表皮細胞の分化が抑制されるとともに、IL-36の発現が誘導されることがわかった。より詳細に検討するために平成28年度には、ケラチン 6のプロモーターにNotch1もしくはNotch2のcDNAにつなげた遺伝子導入マウスの作成を試みた。50以上の受精卵に遺伝子を注入したが、遺伝子導入マウスは作成できず、胎生致死に陥っていると思われた。導入遺伝子のコンストラクトを変更して、薬剤による誘導系の導入が必要と思われた。
以上の結果をふまえ、皮膚潰瘍での再生表皮でNotchの発現が低下するのは、表皮細胞の分化を抑制して細胞の可塑性を高めること、IL-36を誘導して外来からの病原微生物に対して耐性を高めること、という生物学的意義があることが明らかになった。
