Hypotonic stimulation enhances cellular uptake and cytocidal effect of anticancer drug in gastric cancer cells

Outline of Annual Research Achievements

胃癌細胞株を用い、低浸透圧刺激による細胞内PTX取り込み・抗腫瘍作用への増強効果をin vitroで解析した。接着状態での取り込みはプレートリーダーで、浮遊状態での解析は、フローサイトメトリーで行った。両状態で、胃癌細胞株を各種低浸透圧環境下に、Oregon greenで蛍光標識されたPTXで処理したところ、低浸透圧レベルに依存して、細胞内PTX濃度が上昇することを確認した。浸透圧の細胞増殖への影響は浮遊状態で特に顕著であり、低浸透圧刺激+PTX群の細胞増殖が最も抑制された。PTXの膜輸送体の発現を解析したが、浸透圧変化に伴う遺伝子発現変化は認められず、脂質二重層を介する取り込み増強効果が示唆された。
また、ヌードマウス腹膜播種モデルを用い、播種結節へのPTX取り込みをMVX10 MacroView ・蛍光顕微鏡で解析した。浸透圧の低下に伴い、蛍光標識されたPTXの播種結節への取り込みが結節辺縁より上昇することが確認できた。
一方で、胃癌細胞株における低浸透圧殺細胞効果が、K+チャンネル阻害剤により調節性容積減少(RVD)の抑制を介して増強されることを明らかにした。また、癌細胞への温熱刺激における、水輸送体のAutophagyを介する制御機序の解明(Int J Oncol. 2017)や、水輸送体の制御を介してRVDを抑制し、低浸透圧殺細胞効果が増強されることも示した(J Cancer. 2016)。同時に、消化器癌におけるAE1 (Oncotarget. 2017)、NHE1(Oncotarget. 2017)、TRPM7 (Anticancer Res. 2017)等の細胞生理学的因子や、アポトーシス阻害因子(Dig Dis Sci. 2017)の機能解析を行った。更に、癌幹細胞に特異的に発現するイオンチャネルを同定した(J Gastroenterol. 2017)。
