Outline of Final Research Achievements

In this study, leptin sensitivity in hypothalamus after endurance exercise and its mechanism were examined. Male C57BL/6J mice were performed treadmill running for 45 minutes. After that, leptin was injected intraperitoneally. Forty-five minutes later, hypothalamus in mice was collected, and determined the phosphorylation of STAT3 by western blot or immunohistochemistry.
Treadmill running enhanced STAT3 phosphorylation in hypothalamus of the mice by ip injection of leptin. Immunohistochemical staining revealed that treadmill running increased c-Fos expression in one neuropeptidergic neuron in hypothalamus of the mice. Intracelebroventricular injection of receptor antagonist of the neuropeptide inhibited the exercise-induced enhancement of STAT3 phosphorylation in hypothalamus by leptin. Taken together, it is suggested that the enhanced leptin sensitivity after exercise is mediated by activation of the neuropeptidergic neuron by exercise.
