ヒメミカヅキモの性決定遺伝子から迫る、ストレプトファイツ植物の性決定

Outline of Annual Research Achievements

本研究では、－型細胞ゲノムのみに存在し、＋型細胞で特異的に発現する遺伝子群を発現抑制する遺伝子（CpMinus2）を、CpMinus1遺伝子との連鎖を前提に近傍のゲノム情報を精査することで明らかにすることを目的とした。H27年度には、PacBio P6-C4ケミストリーを用いたゲノム解読を行った。結果として、両接合型細胞のゲノム情報のアセンブルが向上したものの、ヒメミカヅキモ用にアルゴリズム設計をした、Illumina short read dataとPacBioデータのハイブリッドアセンブラーの開発に難航しており、CpMinus1遺伝子を含む－型性染色体領域の全貌は未だ解明されず、さらに＋型性染色体領域については、全く明らかにならなかった。
一方で、CpMinus1遺伝子をCpMinus1 native promoter下流に連結し、あらたな＋型細胞形質転換体作出に成功した。これらの株では、CpHSP70 promoterにてCpMinus1遺伝子を発現させた場合に比べて、遥かに高い発現上昇が示され、－型細胞と比べても、その10倍近い発現レベルを示した。これらは顕著に－型化傾向を示したものの、依然として自家生殖能力を示した。これらから、CpMinus2遺伝子、または＋型細胞ゲノムのみに存在し、＋型細胞で特異的に発現する遺伝子群の発現を正に制御する遺伝子（CpPlus1）が存在することが確定的となった。それらをtranscriptomeデータからも探るため、PacBioによるIso-seq解析のためのライブラリー調製を進めた。また、オーストラリアシャジクモ（Chara australis、雌雄異株）についても、同様にライブラリー調製を進めた。

Reason

ゲノムのscaffoldingには、相変わらず難航しているが、PacBio P6-C4ケミストリーデータとIllumina short read dataとのハイブリッドアセンブラーの開発され次第、一気に進むものと思われる。また、仮定に過ぎなかったCpMinus2, もしくはCpPlus1の存在が強く示唆されるようになったことも重要な進展である。ヒメミカヅキモ、オーストラリアシャジクモでのIso-seq解析のためのライブラリー調製が進行しており、総合的に見て、想定の範囲内で進展していると判断した。

Strategy for Future Research Activity

ヒメミカヅキモ用のアルゴリズムを適用したハイブリッドアセンブラーの開発を期待しつつ、並行してIrys BioNanoによるscaffoldingを行う。また、各12系統程度の子孫株からゲノムデータを得つつあり、マッピングにより性染色体領域の絞り込みを進める。CpMinus2またはCpPlus1を特定し、性決定への関与を解析する。

Causes of Carryover

Iso-seq解析にはかなりの消耗品経費が必要となるが、共同研究先（国立遺伝研）のPacBio稼働状況から、2015年度中には解析が難しい状況であったため、2016年度での解析にて使用することとした。
