本研究では、マイナス型細胞ゲノムのみに存在し、プラス型細胞で特異的に発現する遺伝子群を発現抑制する仮想遺伝子CpMinus2を、CpMinus1遺伝子との連鎖するゲノム領域を精査することで明らかにすることを同定することを第一の目的としている。CpMinus1遺伝子破壊株の作出と表現型解析の結果、仮想遺伝子であるCpMinus2の存在する可能性は非常に高まったものの、ヒメミカヅキモの部分的な配列重複などの影響もあり、アセンブルが難航していた。しかし、PacBio P6-C4ケミストリーを用いたゲノム解読データを、バージョンアップしたアセンブラーFalconを用いることにより、プラス株においては、primary contigがN50 416 kbでtotal 318 Mb、associated contigがN50 60 kbでtotal 42 Mbまで改善した。マイナス株の場合も、primary contigがN50 319 kbでtotal 302 Mb、associated contigがN50 58 kbでtotal 34 Mbまで改善した。そのうえで、子孫株の性表現とIlluminaによる解析の結果から、マイナス型特異的なゲノム領域は存在するものの、プラス型特異的なゲノム領域は存在しない可能性が高まった。また、CpMinus1を含むcontig自身は、あまり長く伸びてはいなかったため、その中からCpMinus2の候補配列は得られなかった。
第二の目的であるCpMinus1オルソログ遺伝子をオーストラリアシャジクモから見出し、その性表現への影響を調べることについては、Isoseq解析データを取得した。その中からCpMinus1のホモログはいくつか存在していたが、オルソログとは断定できなかった。
