Outline of Annual Research Achievements

幹細胞研究の発展により、多能性幹細胞(ES細胞、iPS細胞)や組織幹細胞(脂肪組織由来間葉系幹細胞)から様々な組織への分化誘導法が開発され多数の報告がなされている。しかしながらこれまでの分化誘導の報告の大多数が生体外において作成し、細胞療法を行うためのソースとしては有用であるが、臓器障害を有し、移植以外の選択肢の無い多くの患者では細胞療法のみで完全に臓器不全を補完することは極めて困難である。そこで三次元の組織の作成を目指し膵臓の無いマウス体内でのラット膵臓の作成や、膵臓の無いブタに健常なブタの膵臓を作成する報告がなされ、異種の受精卵内にヒトiPS細胞やES細胞を移植し発生させることで異種の体内でヒトの臓器を作成することが有望であると考えられている。マウスES、iPS細胞は異種動物の細胞をキメラ化し、その状態のまま正常に受精卵を発生させ個体を得ること可能であるが、マウスES細胞よりも発生の進んだマウス胚盤用上層幹細胞(EpiSC)はキメラ系性能を持たないことが知られている。そしてヒトES、iPS細胞はマウスES、iPS細胞と性質に大きな違いがあり、マウスEpiSCとより多くの共通点を持つことからキメラ系性能を持たないと考えられている。そこで、本研究では受精卵由来新規マイクロRNAを固定しキメラ形成能を有するヒトiPS細胞の樹立を目指した。
