酸素含有脂質微小粒子による生体内酸素ホメオスターシス維持法開発を目指した基礎研究

Outline of Annual Research Achievements

麻酔科医が遭遇する気道閉塞などによる肺胞換気停止・不能に伴う低酸素血症の治療につながる静脈内投与可能な酸素マイクロバブル製剤の開発のための基礎研究を行う事が本研究申請の目的である。静脈内投与に最適化された脂質二重膜を用いた酸素含有脂質粒子の作出法を確立すること、その有効性を培養細胞・実験小動物レベルで他のバイタルパラメータと共に酸素ホメオスターシスセンサー低酸素誘導性因子1の活性化などの視点から検証することが具体的な目的であった。以下に研究の実施状況を記載する。

#1 静脈内投与に最適化された脂質二重膜を用いた酸素封入微小バブル(LOM)の作出法を確立 - プロトタイプの作出には成功したが研究計画に記載した所期の性質を備えた酸素封入微小バブルの作出には至っていない。
#2 酸素封入微小バブルの有効性を培養細胞を用いた実験系での検証 -細胞内シグナル伝達、遺伝子応答の検討はHIF-1の活性化を指標とした細胞内低酸素状態の検証の実験系を確立して酸素バブルを用いて評価系を確立した。
#3 酸素封入微小バブルの有効性をラットを用いた実験系での検証-プロトタイプのLOMを用いて実験を試みたが所期の結果は得られなかった。
