Outline of Annual Research Achievements |
既存の力学物性を遥かに凌ぐ高強度・高靱性ハイドロゲルが次々と開発されており、その代表格であるダブルネットワーク(DN)ゲルは約90%が水であるにも関わらず、天然軟骨組織に匹敵する力学物性を有している。ウサギを用いたin vivo試験においてDNゲルの低摩擦性を利用した人工軟骨や軟骨組織再生用の足場材料として極めて良好な成果が得られた一方で、その水様性表面のためにあらゆる接着剤は効果的に使用できず、生体内環境下での強固なゲルの接着・固定が大きな課題であった。平成26、27年度では、骨の無機主成分であり骨伝導性を示すハイドロキシアパタイト(HAp)をDNゲル表面に複合させ、ウサギの大腿骨顆部の欠損へ埋植したところ、4週でゲル母材の強度を超える強い接着を達成した。透過型電子顕微鏡像からゲル内部へ骨再生が進展し融合層の形成が確認された。平成28年度ではこの融合層にフォーカスし、定量的な接着強度を評価するための世界初のバイオメカニカル試験系を構築した。比較的広い平面を確保できる頭頂骨に作製した平面欠損部へシート状のHAp/DNゲルを埋植し、2、4、6週後に90°剥離試験を行って接着強度を評価した。その結果、週数を追うごとに接着の確率と接着強度が向上し、2週に比べて6週では約2倍の強度を示した。また、頭頂骨部は大腿骨顆部に比べて骨再生能が低いにも関わらず、十分な接着強度を示し、本接着手法があらゆる骨組織に対応できる汎用性が示された。融合層のオーバーラップ長を見積もるために、同位体ラベル化されたHApを用いた同位体マッピングを現在測定中である。これらの結果を基に、物質透過性の高いDNゲル特有の相互侵入型骨接着の構造及びメカニズムの総括を発表予定である。またこれらの成果に関心が持たれ、2017年1月より企業と実用化に向けた共同研究を締結した。
