抗プリオン活性を有する四重鎖核酸の探索とそれらの作用機構に基づく分子設計

Outline of Annual Research Achievements

プリオン蛋白質の正常型から異常型への構造遷移がプリオン病の原因と考えられている。申請者は正常型プリオン蛋白質に強く結合する12残基のRNA分子(以後R12)が、プリオン蛋白質の構造遷移を抑制できること、すなわち抗プリオン活性を有することを見出した。本研究では、R12の立体構造を基に抗プリオン活性の高い核酸分子を設計し、異常型プリオン蛋白質を産生する細胞を用いてその抗プリオン活性を検証した。その結果、R12以上に抗プリオン活性の高いRNA分子(以後R24)を見出すことに成功し、そのIC50を調べたところ、100 nM程であった。
最終年度では、R24の立体構造とプリオン蛋白質との相互作用様式について、NMR法による解析を目指した。しかし、R24のNMRスペクトルのシグナル本数は、R24の残基数から予想されるシグナル本数よりも遥かに多かった。これはR24の立体構造に多型性があり、各コンホメーションのシグナルが同時に観測されたためであると考えられる。R24を溶解する溶媒や調製方法を検討したが、R24の立体構造を1系にすることはできなかった。そこで、R24の配列の一部を変更あるいは末端残基の付加等によって、単一のコンホメーションで存在するR24変異体の取得を試みた。系統的に設計したR24変異体の中から、各シグナルがよく分離し、かつNMRスペクトルのシグナル本数が残基数から予想されるシグナル本数とほぼ一致するR24変異体を見出すことに成功した。
