Outline of Final Research Achievements

We attempted to generate mesothelin-specific cytotoxic T lymphocytes (CTLs) for developing novel immunotherapy for treating refractory pediatric acute myeloid leukemia. We cultured monocytes from peripheral blood mononuclear cells in a cytokine mix for 1 week so that monocytes could differentiate into dendritic cells. Furthermore, we attempted to generate mesothelin-specific CTLs after stimulating T lymphocytes with mesothelin-peptide-pulsed dendritic cells and a cytokine mix. Although we were unable to effectively generate mesothelin-specific CTLs, we found variable specificity in the generated mesothelin-specific CTLs. We believe our method needs further improvements before it can be applied clinically.
