頭頸部癌におけるcyclin D1標的sgRNAによる新規遺伝子標的治療の開発

Outline of Annual Research Achievements

TRUE gene silencing法を用いCCND1を標的とした数種のsgRNAを設計し，頭頸部癌細胞培養系においてこれらの導入によるCCND1発現抑制および増殖抑制作用を調べることを目的として下記の実験を行い研究実績を得た.
【方法】Human CCND1 mRNAの二次構造を解析し，5’-half-tRNA (HT)型，heptamer (H)型およびlinear (L)型のsgRNA (HT1-6, H1-6, L1-6) を設計した．これらの18種類のsgRNAについて，2’- O-methyl化したRNAを化学合成した．ヒト頭頸部扁平上皮癌細胞株であるHSC-2およびHSC-3細胞を用い，カナマイシン及び 10％ FBSを含むRPMI培地にて培養した．細胞を培養用dishに播種し，24時間後lipofectamine2000添加もしくは無添加で種々のsgRNAをトランスフェクションした．引き続き細胞を培養後，total RNAを抽出し，リアルタイムPCR法によりCCND1 mRNA量を定量した．また，Western blot法によりCCND1のタンパク量を測定した．
【結果】HSC-2およびHSC-3細胞いずれの細胞でも，CCND1 mRNA量は，各種sgRNAの導入によって20-70%のレベルに低下した．これらのsgRNAのCCND1発現抑制効果は，lipofectamineを用いないで培地に加えただけの場合でも認められた．設計したsgRNAの標的部位によって抑制効果には差があり，HT型では，HT-2, HT-5による抑制効果が大きかった．sgRNAのHT型，H型およびL型の違いはCCND1発現抑制活性には大きな差異は認められなかった．Western blotによる検討から，sgRNAの導入によってCCND1のタンパク質レベルも低下した．
【研究実績】以上の研究から，新しい遺伝子発現制御法であるTRUE gene silencing法を用い， 適切に設計されたsgRNAの導入によって，頭頸部癌細胞であるHSC-2およびHSC-3細胞においてCCND1の発現を抑制させることが明らかになった

Strategy for Future Research Activity

sgRNAをトランスフェクト後、シスプラチン（CDDP）添加し、生細胞数をMTT法，Capase3/7活性をCaspase-Glo 3/7 Assay Systemsを用いて測定することで、遺伝子発現及びタンパク発現のみならず生細胞数が減少していることを確認しsgRNAが有する抗がん効果を検証する,
