Research Abstract

On two-dimensional TLC, each tissue of Aplysia showed a pattern of phosphonoglycosphingolipids characteristic to the tissue. FGL-IIa and FGL-IIb were specific to the nerve fiber and FGL-V (G-13) was seemed to be specifically concentrated in the nervous tissues. Three novel phosphonoglycosphingolipids (FLG-IIa, FGL-IIb and FGL-V) were isolated from the nervous tissues of Aplysia kurodai, using three systems of silicic acid column chromatography and characterized. FGL-IIa was monophosphonoglycosphingolipid having an oligosaccharide chain consisting of glucose, galactose and galactosamine. FGL-IIb was also monophosphonoglycosphingolipid having fucose, glucose, galactose and galactosamine as an oligosaccharide moiety. FGL-V (G-13) seemed to be diphosphonoglycosphingolipid having an oligosaccharide chain consisting of glucose, galactose, galactosamine and 4-0-methylglucosamine.
Antisera against FGL-IIa and FGL-IIb mixture were raised in rabbits. Two-dimensional thin-layer chromatography combined with immunoautoradiography using the antisera and <^(125)I> -Ig,F(ab')2, revealed nine immunoreactive glycolipids in extracts from the nerve fiber. By immunohistochemistry with the antisera and HRP-labelled antirabbit IgG, distinct and strong staining was observed in the nerovus tissue. Nerve fibers in the skin and muscle tissue were also stained.
