Research Abstract |
The pathogenesis of minimal change nephrotic syndrome(MCNS) is still unclear. Recently there were several reports that T cell dysfunction was observed, associated with the onset of MCNS and a kind of "lymphokine" might cause abnormal increase in the glomerular vascular permeability. In this study, we investigated the characteristics of the factors which cause the glomerular vascular permeability by the production of lymphokines using human T cell hybridoma. The lymphocytes from patients with MCNS were hybridized with CCRF-HSB2 cells, by polyethyleneglycole method. The factors which produce urinary protein was assayed by the iv injection of the culture supernatants into rats and examined the effects of lymphokines on IL-II dependent MTH mouse cell line, ConA-induced lymphocyte blast formation, grouth of cell lines from hematogenous organ and carcinoma. We observed factor(s) which caused significant urinary proteins and also enhanced normal lymphocye grouth (more than 10 times) in ConA-induced lymphocte blast formation. The supernatants also showed cytotoxic effect on carcinoma cell lines. However, during cloning, the activities of the factor decreased. We supeculated that thevvascular permeability factor and T cell activating factor might be a lymphokine which T cell produced in patients with MCNS, and play an important role in pathogenesis of MCNS.
