Research Abstract

ヘルペスウイルス科のウイルス、中でも単純ヘルペスウイルス(HSV),水痘帯状疱疹ウイルス(VZV),サイトメガロウイルス(CMV)及びヒトヘルペスウイルスNo.6(HHVー6)の潜伏感染とその再活性の機構及びその抑制について研究を以下のように行なった。(1)SCIDマウスを用いてHSVに対する感染防御機構の解析を行なった。また、SCIDマウス及びヌ-ドマウスなど免疫不全宿主においてもHSV接種では潜伏感染が成立し、妊娠によって再活性化がおこることを見出した(森)。(2)HSVは1型と2型があるが両者の型別を行なうための基礎的条件の設定の研究を行なった(栗村)。(3)HSV1型の株間における再活性化可能な潜伏感染能の比較を行なった(新居)。(4)ヒトにおけるHSVの感染における神経組織の関与について詳細な解析を行なった(倉田)。(5)HHV6及びVZVの潜伏感染及びその再活性化のあり方をin vivo,in vitroの両面から調べると共に、その感染様式を明らかにするため、咽頭ぬぐい液などからPCR法を用いてウイルスDNAの検出を試みた。その結果突発性発疹患者の急性期にはCD4^+リンパ球及びマクロファ-ジの両者にHHVー6 DNAが証明されることを明らかにした。また回復期にはマクロファ-ジのみからHSVー6 DNAが証明されることを明らかにした。(高橋)。(6)ヒトCMVの潜伏臓器、潜伏様式を明らかにするためにPCRを応用した(古川)。(7)妊娠経過にともない子宮頸管からCMVが排泄されるようになるが、この場合補体要求性中和抗体の動きを調べ、再活性化への影響について検討した(田中)。(8)シェ-グレン症候群におけるCMV感染の意義について自己抗体の面から解析を行なった(清水)。(9)弱毒変異マウスCMVを用いてマウスにおける致死感染及び持続感染の遺伝子レベルでの解析を行なった(南嶋)。
