| Research Abstract |
妊娠ラット卵巣粗膜分画に対する^<125>Iーinsulin,^<125>IーIGFーIの結合を検討したところ、それぞれのレセプタ-の存在が確認され、Irsulinレセプタ-はKd=0.031nM,Bmax=1.24fmol/mg tissue,IGFーIレセプタ-はKd=0.024nM,Bmax=0.338fmol/mg tissueであった。Insulinレセプタ-に対する結合の強さはInsulin,IGFーII,Iの順でIGFーIレセプタ-に関してはIGFーI,II,Insulinの順だった。妊娠ラット卵巣細胞をInsulin,IGFーI,IIとともに培養したところ,CAMP Phosphodiesteruse阻害剤であるIBMX併用時に、P^4,20αーOHーP^4,CAMPが増加した。この作用はIBMX非存在時には出にくかったので、Irsulin等により、Phosphodiesteruse活性が亢進していることが考えられた。そこで、^3HーCAMPを基質として卵巣粗膜分画のPhosphodiesteruse活性を測定したところ、機序は不明だったが、5分間のインキュベ-トでInsulinとIGFsが酵素活性を亢進させることが判明した。
