Research Abstract

The endolymphatic sac and cochlea including spiral modiolar vein were investigated in the guinea pig inoculated Guinea Pig Cytomegalovirus (GPCMV) and psoralen-ultraviolet inactivated GPCMV into the cochlea of seronegative and seropositive animal.
Histopathological including immunoelectron microscopic study and electophysiological investigation were carried out at day 2, 4, 6, 8, to elucidate the possible immunological role of endolymphatic sac and cochlea in inner ear viral infection.
The summary of results are as follows ;
1) Hearing loss was found in the seronegative group whereas hearing was protected in the seropositive and control group.
2) It was recognized that inflammatory cells infiltration via collecting venule and spiral modiolar vein from systemic circulation actually occurs in the GPCMV inoculation to the cochlea of seronegative group.
3) No inflammatory cells infiltration was recognized in the cochlea including spiral modiolar vein of seropositive and control group.
4) Only slight inflammatory cells infiltration was found in the endolymphatic sac in this system. But endolymphatic sac reacts immunologically with direct inoculation of GPCMV into the endolymphatic sac.
5) There must be 2 system to react to the stimulation of virus inoculation to the cochlea. Namely, spiral modiolar vein of the cochlea and endolymphatic sac. The degree how these area reacts to the viral infection in the inner ear is still controversial and wait for further precise time cours e study in this particular area.
