Research Abstract |
Recent investigations of the interaction between endothelial cells and vascular smooth muscle clls(VSMC) have disclosed mutual regulatory mechanism, utilizing humoral factors, between the two cells. Such regulatory humoral factors include endothelin, EDRF and prostaglandins. Present study has, in this context, focused on the interrelationship between the two cells. To pursue this, I assessed the effect of endothlium-derived hormone(ET-1) on cation transport of VSMC.In addition, I attempted a new culture system, co-culture of VSMC and endothelial cells. The 1st part of the study showed that ET-1 stimulates Na-K pump activity, Na-H antiport and Na-K contransport. ET-1, per se, also stimulates Ca influx, resulting in an increase in cytosolic free Ca concentration of VSMC.Knowing that a nitro-based agent may mimic the effect of EDRF, the 2nd part of this study described the effect of nitro-based pharmacological agent on cation transport s stems of VSMC, which showed an inhibitory action of NO on the proliferation of VSMC. Because of time-c nsuming technical difficulties, the co-culture experiment could not be completed within the study's 2-year limit and thus was terminated without results.
