Research Abstract

研究代表者はこれまでに低温赤外分光法を用いて、分子ポンプであるバクテリオロドプシンに結合した内部結合水の性質を明らかにしてきた。本研究はこれを蛋白質が実際に機能する室温でも起こるのかどうか明らかにするため、室温での時間分解赤外分光による解析を行うことを計画した。
研究分担者は、研究代表者の研究室に既設のステップスキャン時間分解フーリエ変換赤外分光装置およびナノ秒レーザーを用いて、時間分解赤外分解計測系を構築した。得られたデータを参考にして、分担者が得意とする数学的手法を使った原著論文をApplied spectroscopy誌に発表することができた。その解析をさらに深めた次の論文も最近、Applied spectroscopy誌にアクセプトされ、2007年に出版される予定である。
さらに、バクテリオロドプシンの水和フィルムに対して水分子の信号を得ることに成功し、低温分光法によって得た水の信号と異なる信号を得た。この成果は、低温と室温での光反応ダイナミクスの違いを直接的に示すデータである。現在、原著論文を作成しているところであり、分担者はこの成果を中国での国際会議で招待講演として発表した(2006年11月)。
研究代表者は上記、示した研究成果の他にも、研究発表に記した種々の成果を、主に赤外分光法により得ることができた。
