Research Abstract

Achaeoglobus fulgidus由来のアラニルtRNA合成酵素の結晶化と予備的なX線回折実験に成功した.またAlaRSの校正反応ドメインのホモログタンパク質であるAlaXのX線結晶構造を2.7A分解能で決定した.AlaXはNドメインとCドメインの2つからなっていた.Cドメインに亜鉛を結合した校正反応活性部位があり,Nドメイン中の保存されたグリシンに富むループ(glycine-richループ)がその活性部位近くに位置していた.
ホスホセリルtRNA合成酵素(SepRS)はホスホセリン(Sep)をtRNA^<Cys>に結合させる.A.fulgidus SepRS・tRNA^<Cys>・ホスホセリンの3者複合体のX線結晶構造を2.6Å分解能で決定した.SepRSはN端伸長領域,アミノアシル化活性ドメイン,挿入ドメイン,C端のアンチコドン結合ドメインの4つの構造要素からなっていた.SepRSは結晶中でホモ4量体を形成し2分子のtRNA^<Cys>を結合していた.ホスホセリン側鎖のリン酸基がアミノアシル化活性ドメイン中で強固に認識されていた.C端のアンチコドン結合ドメインがtRNA^<Cys>のアンチコドンループを認識していた.構造に基づいてアンチコドン認識部位に変異を導入し,サプレッサーtRNA(tRNA^<Opal>およびtRNA^<Amber>)を認識する改変SepRSを創出した.これはコドン表を拡張しホスホセリンを加え,タンパク質翻訳時にホスホセリンを部位特異的に導入する技術に利用可能である.改変SepRS・tRNA^<Opal>・ホスホセリン3者複合体および改変SepRS・tRNA^<Amber>・ボスホセリン3者複合体のX線結晶構造をそれぞれ3.2および3.3A分解能で決定し,改変SepRSによるナンセンスアンチコドンの認識機構を解明した.
